Isolation and Characterization of Immunodiagnostic Antigen from Strongylus vulgaris Infecting Horses
نویسندگان
چکیده
Isolation of immunodiagnostic fraction from Strongylus vulgaris adult worms was described by CNBr-Sepharose 4B affinity column chromatography. The isolation process resulted in a fraction with 2161.5 fold increase in binding activity compared to its crude extract. Characterization of the isolated fraction by SDS polyacrylamide gel electrophoresis and isoelectric focusing showed that the fraction consists of only two bands of 39 and 31 KDa with isoelectric points of 6.8 and 6.7. Comparative evaluation of the immunogenic binding activities of the crude extract, unbound and bound fractions by ELISA proved the potency of the bound fraction over the other two antigens. Diagnosis of S. vulgaris infection in horses by ELISA in which the bound fraction was utilized, recorded high infection percentage (73.7) as compared with the parasitological examination (27.6). The isolated fraction in the current study could be utilized successfully in the diagnosis of S. vulgaris infection using ELISA, particularly in the prepatent period.

Cranial Mesenteric Arterial Obstruction Due To Strongylus vulgaris Larvae in a Donkey (Equus asinus)
Arteritis due to Strongylus vulgaris is a well-known cause of colic in horses and donkeys. The current report describes a fatal incidence of arterial obstruction in cranial mesenteric artery caused by S. vulgaris infection in an adult donkey in which anthelmintic treatment was not regularly administered. Necropsy findings of the abdominal cavity revealed a complete cranial mesenteric arterial o...
متن کاملChanges in Serum Strongylus Vulgaris-Specific Antibody Concentrations in Response to Anthelmintic Treatment of Experimentally Infected Foals
Strongylus vulgaris is the most pathogenic nematode parasite of horses. Its extensive migration in the mesenteric blood vessels can lead to life-threatening intestinal infarctions. Recent work has shown that this parasite is still identified among managed horse populations. A serum enzyme-linked immunosorbent assay (ELISA) has been developed for the detection of migrating larvae of S. vulgaris....
متن کاملDetection of Strongylus vulgaris in equine faecal samples by real-time PCR and larval culture – method comparison and occurrence assessment
BACKGROUND Strongylus vulgaris has become a rare parasite in Germany during the past 50 years due to the practice of frequent prophylactic anthelmintic therapy. To date, the emerging development of resistance in Cyathostominae and Parascaris spp. to numerous equine anthelmintics has changed deworming management and the frequency of anthelmintic usage. In this regard, reliable detection of paras...
متن کاملAn evidence-based approach to equine parasite control: It ain’t the 60s anymore
Most veterinarians continue to recommend anthelmintic treatment programmes for horses that derive from knowledge and concepts more than 40 years old. However, much has changed since these recommendations were first introduced and current approaches routinely fail to provide optimal or even adequate levels of parasite control. There are many reasons for this. Recent studies demonstrate that anth...
متن کاملRole of Intrinsic and Extrinsic Epidemiological Factors on Strongylosis in Horses
A total of 133 (65.51%) faecal samples were found positive for various helminths and most frequently strongyle infections were diagnosed with an infection rate of 58.5% (117/200) in a year long study to asses various epidemiological factors influencing the gastro-intestinal nematodes of horses. Significant difference in the prevalence of various gastrointestinal helminths was detected in the st...
